The atom, the molecule, and the covalent organic framework

Christian S. DiercksORCID; Omar M. YaghiORCID

A framework for molecular assembly
Covalent molecular frameworks are crystalline microporous materials assembled from organic molecules through strong covalent bonds in a process termed reticular synthesis. Diercks and Yaghi review developments in this area, noting the parallels between framework assembly and the covalent assembly of atoms into molecules, as described just over a century ago by Lewis. Emerging challenges include functionalization of existing frameworks and the creation of flexible materials through the design of woven structures.
Science , this issue p. eaal1585

Structure of a eukaryotic voltage-gated sodium channel at near-atomic resolution

Huaizong ShenORCID; Qiang Zhou; Xiaojing Pan; Zhangqiang LiORCID; Jianping WuORCID; Nieng YanORCID

Navigating regulated cell excitation
Voltage-gated sodium (Na v ) channels respond to a change in voltage potential by allowing sodium ions to move into cells, thus initiating electrical signaling. Mutations in Na v channels cause neurological and cardiovascular disorders, making the channels important therapeutic targets. Shen et al. determined a high-resolution structure of a Na v channel from the American cockroach by electron microscopy. The structure affords insight into voltage sensing and ion permeability and provides a foundation for understanding function and disease mechanism of Na v and the related Ca v ion channels.
Science , this issue p. eaal4326

Evolving policy with science

R. Alta Charo; Richard O. Hynes

Albert Schweitzer, physician and Nobel laureate, wrote: “The first step in the evolution of ethics is a sense of solidarity with other human beings.” The evolution of U.S. policy governing heritable human germline modification reflects this observation, and a “Human Genome Editing” consensus study committee of the U.S. National Academy of Sciences and National Academy of Medicine (NAM) concluded last month that clinical trials for heritable editing, under strict regulations, may eventually be permissible for parents hoping to prevent serious diseases in their children.
