Agents that increase natural protective mechanisms have been proposed for prevention and treatment of intramammary infections. Staphylococcus aureus is a major pathogen causing primarily sub clinical chronic mastitis, which responds poorly to antibiotic therapy. The objectives of this study were to describe the effects of a single intramammary infusion of a lipopolysaccharide-based biological response modifier (BRM) on a) cellular proliferation, b) cellular death mechanisms, c) unspecific immune response, d) cellular infiltrating cells quantification and e) growth factors expression, in uninfected and S. aureus-infected bovine mammary glands during involution. In conclusion, BRM treatment in uninfected mammary glands induced macrophages infiltration, increased apoptogenic proteases staining and apoptosis of both epithelial and stromal mammary cells. On the other hand, BRM treatment in S. aureus-infected mammary glands induced an increase of TNF-alfa, infiltrating macrophages, pro-apoptotic proteins (Bax), apoptogenic proteases and apoptosis, neither causing deleterious effects on mammary tissue nor inhibiting mammary cell proliferation. In adition, presence of S. aureus IMI during mammary involution resulted in an increase in VEGF and IGF-I expression that was not affected by BRM treatment.
